生防菌GHt-q6对黄瓜枯萎病菌的抑制作用

摘要 设施黄瓜连作现象造成枯萎病的严重发生,影响黄瓜的产量和品质。试验采用平板对峙法、菌落直径生长抑制法和孢子萌发法等测定了生防菌株GHt-q6对黄瓜枯萎病菌的抑制作用。结果表明,生防菌GHt-q6对黄瓜专化型尖孢镰刀菌、辣椒专化型尖孢镰刀菌、禾谷镰刀菌和半裸镰刀菌4种病原菌均有良好的抑制作用;对峙培养中,生防菌GHt-q6会破坏黄瓜枯萎病菌丝生长,菌丝发生畸形、膨大、断裂;黄瓜枯萎病菌丝在含浓度为5%、10%和25%生防菌GHt-q6发酵液平板上的抑制率分别为43.36%、63.87%和78.38%;黄瓜枯萎病菌孢子悬浮液与生防菌GHt-q6发酵液体积比分别为1∶1、2∶1、3∶1时,培养6 h孢子萌发抑制率均为100%,12 h后分别为58.45%、38.38%、24.27%,萌发孢子产生的芽管膨大,随着发酵液浓度越高,芽管越短,24 h后孢子变形成泡囊状,芽管畸形。生防菌GHt-q6对致病镰刀菌有较好的抑制作用,具有开发为生物农药的潜力。 Continuous cropping of greenhouse cucumber caused serious occurrence of Fusarium wilt,which affected the yield and quality of cucumber.The inhibition effect of biocontrol bacteria GHt-q6 on cucumber Fusarium wilt was determined by plate confrontation method,strain diameter growth inhibition method and spore germination method.The results showed that the biocontrol bacteria GHt-q6 had a good inhibitory effect on four kinds pathogenic bacteria of Fusarium oxysporum f.sp.cucumerinum,Fusarium oxysporum f.sp.capsicum,Fusarium graminearum and Fusarium semitectum.In the confrontation culture,the biocontrol bacteria GHt-q6 destroyed the growth of cucumber Fusarium wilt hyphae,the hyphae were deformed,swelled and fractured.The inhibition rates of cucumber Fusarium wilt hyphae on the fermentation broth plate containing 5%,10%and 25%of the biocontrol bacteria GHt-q6 were 43.36%,63.87%and 78.38%,respectively.When the volume ratio of cucumber Fusarium wilt spore suspension and biocontrol bacteria GHt-q6 fermentation broth were 1∶1,2∶1 and 3∶1,culture for 6 h the inhibition rates of spore germination were 100%,and after 12 h they were 58.45%,38.38%and 24.27%,respectively.The germ tubes produced by germinated spores were swelled,with the higher the concentration of the fermentation broth,the shorter the germ tube.After 24 h,the spores transformed into vesicles and the germ tubes were deformed.The biocontrol bacteria GHt-q6 had a good inhibitory effect on the pathogenic Fusarium,and had the potential to be developed as a biological pesticide.
